What is the "4 afm"? I used Comp R lifters in my last LS build, would those be a good choice in the 5.3?

Four of the eight lifters are AFM (active fuel management) style.

OK, besides the crank sensor box, I learned that my engine ('07 5.3) has the cam sensor on the front cover by the crank pulley. Other then the wire/plug that might be too short for the front cam sensor (older style has the sensor in the rear), can I just extend that plug without any issues?

Thats what I would do, cut the wire, solder some quality wire in there to extend it to the front, no cheap wire. Use wires you've stripped out of the harness to extend the leads. Be sure to shrink wrap the connections afterwards.
